Antitope announces the launch of new Composite CHO technology for development of manufacturing cell lines suitable for biologics
Antitope today announced the launch of Composite CHO technology for the generation of CHO cell lines suitable for cGMP manufacture of recombinant antibodies and proteins. The Composite CHO technology has been developed for direct transfer of genes into CHO cells pre-adapted to grow in serum-free medium. As a result, CHO manufacturing cell lines are now available in less than 6 months and can be transferred directly to a cGMP manufacturing facility for manufacture of proteins for clinical trials. When combined with Antitope’s Composite Human Antibody technology, CHO cell lines producing high levels of fully humanised antibodies can be delivered within 12 months of supply of a starting hybridoma or gene sequences.
Dr Tim Jones, Antitope’s Director of Research, said “We are pleased to announce the launch of Composite CHO technology for high level expression of recombinant proteins in mammalian CHO cells. This has resulted from several years of research where we have carefully analysed and optimised all stages of gene transfer and expression by a multi-parametric process. This research has now provided a robust technology which achieves much higher efficiency of gene delivery resulting in higher copy number and higher secretion of recombinant proteins than previously achieved.”
The Composite CHO technology has been tested by Antitope in the last year alongside standard CHO expression technology and an alternative technology from a partner. In this side-by-side comparison, the Composite CHO technology resulted in greatly enhanced productivity and stability than the standard and alternative CHO technologies. As a result, several CHO cell lines with high productivity levels for fully humanised antibodies have been transferred to cGMP manufacturing facilities resulting in successful production of clinical batches.
About Antitope:
Antitope was formed in 2004 by scientists from Biovation/Merck KGaA. The company has established its own proprietary protein engineering and immunogenicity testing technologies and has established a large international client base. Antitope now provides Composite CHO technology as a service to biopharmaceutical companies together with its range of antibody/protein engineering and immunogenicity testing technologies. These include Composite Human Antibody technology for generation of therapeutic antibodies devoid of CD4+ T cell epitopes, Composite Protein technology for generation of therapeutic proteins, iTope/TCED technologies for in silico analysis for immunogenicity potential, and EpiScreen technology for preclinical prediction of clinical immunogenicity.
