Manufacturing Cell Lines
Antitope routinely generates high expressing cell lines using mammalian cells for manufacturing and commercial production of recombinant proteins such as Composite Human Antibodies™ and Composite Proteins™. Standard host cell lines are CHO (Chinese Hamster Ovary) and NS0 (mouse myeloma). Antitope has developed a proprietary expression system (pANT™ Vector System) to enhance expression levels as follows;
- pANT™ Vector System includes a UTR (untranslated terminal repeat) discovered at Antitope to enhance transcription of the gene encoded downstream of this UTR
- for expression of recombinant antibodies, heavy and light chain V region genes are cloned into a single small sized expression vector that allows more efficient transfection of mammalian cells
- pANT™ Vector System contains a modified dhfr gene allowing for rapid amplification of vector copy numbers in mammalian cells by methotrexate selection
Manufacturing cell lines are grown in suspension and produce high yields of recombinant proteins in chemically defined medium. These manufacturing cell lines have been accepted by CMO's and manufacturers worldwide to produce cGMP material.
